Selective Medium for Staphylococci & Streptococci Isolation
This PEA Agar Base requires supplementation with blood and β-phenylethanol to create a selective environment that suppresses Gram-negative bacteria while promoting Gram-positive cocci growth.

⚠️ Critical Preparation:
△ Must add 5-7% defibrinated sheep blood
△ Requires 0.25g/100ml β-phenylethanol
△ For clinical microbiology use only

▶ Base Formula (per liter):
Pancreatic Digest of Casein 15.0g
Enzymatic Digest of Soybean Meal 5.0g
NaCl 5.0g | Agar 15.0g
▶ pH: 7.3±0.2 (25°C)
▶ Preparation:

  1. 40g/L → boil → sterilize at 121°C/15min

  2. Cool 50-55°C → add 50ml sheep blood + 2.5g β-phenylethanol per liter

  • Isolation of Staphylococcus aureus from mixed flora

  • Recovery of Enterococcus species

  • Wound/pus specimen processing
