Skimmed Milk Sucrose Peptone Base is designed for identifying pathogenic Aeromonas hydrophila strains through protease activity detection, a key virulence factor marker.

Key Application:

Supplement Requirement:

– Add 1 vial 20% Skimmed Milk Solution per 100ml medium

Diagnostic Principle:

– Pathogenic strains produce proteases

– Clearance zones around colonies indicate protease activity

– White opaque background enhances visibility

Protocol:

1️⃣ Suspend 36.5g in 1L water

2️⃣ Boil ≥1 minute

3️⃣ Autoclave at 121°C for 15 minutes

4️⃣ Cool to 50-55°C

5️⃣ Add 1 skimmed milk vial per 100ml

6️⃣ Mix gently → Pour plates

Result Interpretation:

– Positive (pathogenic): Clear hydrolysis zones

– Negative: No clearance
