Composition (per liter)

• Tryptone – 20.0g

• Dextrose – 1.0g

• Monopotassium phosphate – 2.0g

• Dipotassium phosphate – 2.0g

• Sodium Chloride – 5.0g

• Tween 80 – 1.5g

• pH 7.0 ± 0.2 (25°C)

Preparation Instructions

1. Dissolve 31.5g of Shigella Broth Base in 1 L of distilled or deionized water with gentle heating.

2. Dispense 225ml per bottle or tube.

3. Sterilize at 121°C for 15 minutes.

4. Cool to 50–55°C.

5. Aseptically add 1 vial (0.125mg) of Novobiocin solution per 225ml, mix thoroughly, and use immediately.
