Composition (per liter)
- Peptone: 10.0g
- Dextrose (Glucose): 40.0g
- TTC (Triphenyl Tetrazolium Chloride): 0.01g

Final pH: 5.6 ± 0.2 at 25°C
Instructions for Preparation
- Suspend 65.0g of the powder in 1 liter of distilled or deionized water.
- Heat and boil to dissolve the medium completely.
- Dispense into final containers.
- Sterilize by autoclaving at 121°C for 15 minutes or 115°C for 30 minutes.
- After sterilization, gently swirl the vessel to prevent agar sedimentation.
- Optional: For enhanced bacterial inhibition, cool the medium to approx. 50°C and aseptically add 1 vial of chloramphenicol (10mg) per 100ml of medium before pouring plates.
