This Sabouraud Dextrose Agar (SDA) is the standard, most widely used culture medium for the isolation, cultivation, and maintenance of pathogenic and non-pathogenic fungi and yeasts. It is a fundamental medium in clinical, food, and pharmaceutical microbiology laboratories worldwide.

This formulation features a specialized 1:1 mixture of animal tissue peptone and pancreatic digest of casein, providing a rich and balanced nitrogen source to support the growth of a wide variety of fungal species, including dermatophytes. The high glucose concentration and acidic pH of 5.6 create selective conditions that inhibit many contaminating bacteria while promoting optimal fungal development.

Instructions for Preparation

  1. Suspend 65.0g of the powder in 1 liter of distilled or deionized water.
  2. Heat and boil to dissolve the medium completely.
  3. Dispense into final containers.
  4. Sterilize by autoclaving at 121°C for 15 minutes or 115°C for 30 minutes.
  5. After sterilization, gently swirl the vessel to prevent agar sedimentation before the medium solidifies.
  6. Cool and pour into plates or prepare slants as required.
