RPF Agar Medium Base is a microbiological culture medium designed for the detection and differentiation of coagulase-positive Staphylococcus species. The medium provides nutrients that support bacterial growth and allows the identification of characteristic reactions when supplemented with rabbit plasma, bovine fibrinogen, potassium tellurite, and trypsin inhibitor.

Preparation Instructions
- Weigh 58 g of the medium powder.
- Dissolve in 900 mL of distilled water with heating until completely dissolved.
- Dispense and sterilize at 121°C for 15 minutes.
- After sterilization, mix well to prevent agar sedimentation and maintain in a 48°C water bath.
- Under aseptic conditions, add the following to each 90 mL of medium:
- 2.5 mg sterile potassium tellurite
- 2.5 mL rabbit plasma
- 7.5 mL bovine fibrinogen solution
- 2.5 mL trypsin inhibitor
- Mix gently and pour into sterile Petri dishes.
Preparation of Supplement Solutions
Bovine Fibrinogen Solution
Dissolve 5 g bovine fibrinogen in 100 mL distilled water and sterilize using 0.22 μm membrane filtration.
Trypsin Inhibitor Solution
Dissolve 30 mg trypsin inhibitor in 30 mL distilled water and sterilize using 0.22 μm membrane filtration.
