The Modified Starkey’s Medium C with 2.5% NaCl is designed for the growth of Desulfobacter and Desulfovibrio species, as well as other sulfate-reducing bacteria. The addition of 2.5% NaCl enhances its utility for more specific environments and is optimized for sulfur reduction studies. It includes 35% Sodium Lactate Solution to further stimulate growth and ensure the desired microbial activity.

Formulation (Per Liter of Medium):

  • NaCl (Sodium Chloride): 25.0g

  • Yeast Extract: 1.0g

  • Potassium Dihydrogen Phosphate (KH₂PO₄): 0.5g

  • Ammonium Chloride (NH₄Cl): 1.0g

  • Sodium Sulfate (Na₂SO₄): 1.0g

  • Calcium Chloride Dihydrate (CaCl₂·2H₂O): 0.1g

  • Magnesium Sulfate Heptahydrate (MgSO₄·7H₂O): 2.0g

  • L-Cysteine Hydrochloride (L-Cysteine·HCl): 0.75g

  • Ferrous Ammonium Sulfate (Fe(NH₄)₂(SO₄)₂): 0.5g

Usage Instructions:

  1. Dissolve 31.85g of Modified Starkey’s Medium C with 2.5% NaCl in 1L of distilled or deionized water.

  2. Heat and stir the solution until fully dissolved.

  3. Sterilize by autoclaving at 121°C for 15 minutes.

  4. After sterilization, cool the medium to room temperature (25°C).

  5. Add 1 vial of 35% Sodium Lactate Solution (contains 0.35g sodium lactate per vial) to every 100ml of the medium.

  6. Adjust the pH to 7.4 at 25°C using sterile Sodium Carbonate Solution or diluted Hydrochloric Acid.

  7. Mix well, and the medium is ready for use.
