Geobacter Medium (ATCC Medium 1957) is formulated as a basal medium for the cultivation of Geobacter species. This medium provides essential nutrients for microbial growth and is used with fumarate (disodium salt) supplementation for effective cultivation under anaerobic conditions.

Specifications:

Formula (per liter):

  • NH₄Cl – 1.5 g

  • NaH₂PO₄ – 0.6 g

  • KCl – 0.1 g

  • NaHCO₃ – 2.5 g

  • Sodium acetate – 0.82 g

  • Wolfe’s Vitamin Solution – 10 ml

  • Modified Wolfe’s Minerals – 10 ml

Preparation Instructions:

  1. Dissolve 5.59 g of medium in 900 ml of distilled or deionized water.

  2. Warm gently until completely dissolved.

  3. Sparge with N₂–CO₂ (80:20) to remove dissolved oxygen.

  4. Dispense into suitable containers and sterilize by autoclaving at 121℃ for 15 minutes.

  5. Cool rapidly to below 50℃.

  6. Under aseptic conditions, add 2 vials of fumaric acid disodium salt solution per 90 ml medium.

  7. Adjust pH to 6.8 (at 25℃) using sterile acid/base solution.
