When supplemented with defibrinated sheep blood, the medium undergoes controlled heating to release growth factors (X and V factors) from red blood cells, producing the characteristic chocolate-brown appearance essential for optimal bacterial recovery.

Medium Composition (per liter)
Peptone: 15.0 g
Beef Extract: 3.0 g
Yeast Extract: 4.0 g
Maize Starch: 1.0 g
Sodium Chloride: 5.0 g
Agar: 15.0 g
Final pH: 7.2 ± 0.2 (25℃)
Preparation Instructions
Suspend 43.0 g of the medium in 1 L distilled or deionized water.
Heat to boiling until fully dissolved.
Dispense into containers and autoclave at 121℃ for 15 minutes.
After sterilization, maintain at ~90℃ and aseptically add 5–10% defibrinated sheep blood.
Mix gently and continue heating with shaking until hemolysis occurs and the medium turns chocolate-brown.
Cool to ~50℃, mix thoroughly, and pour plates.
