Medium Composition (per liter)

  • Peptone: 15.0 g

  • Beef Extract: 3.0 g

  • Yeast Extract: 4.0 g

  • Maize Starch: 1.0 g

  • Sodium Chloride: 5.0 g

  • Agar: 15.0 g

  • Final pH: 7.2 ± 0.2 (25℃)

Preparation Instructions

  1. Suspend 43.0 g of the medium in 1 L distilled or deionized water.

  2. Heat to boiling until fully dissolved.

  3. Dispense into containers and autoclave at 121℃ for 15 minutes.

  4. After sterilization, maintain at ~90℃ and aseptically add 5–10% defibrinated sheep blood.

  5. Mix gently and continue heating with shaking until hemolysis occurs and the medium turns chocolate-brown.

  6. Cool to ~50℃, mix thoroughly, and pour plates.
