Blood Agar Base for Microbiological Culture

🧾 Medium Composition (per liter)

• Meat Extract – 10.0 g

• Peptone – 10.0 g

• Sodium Chloride – 5.0 g

• Agar – 15.0 g

⚙️ Directions for Use

1. Suspend 40.0 g of the medium in 1 L of distilled or deionized water.

2. Heat with agitation and boil until completely dissolved.

3. Dispense as required and sterilize by autoclaving at 121 °C for 15 minutes.

4. Mix well after sterilization to prevent agar sedimentation.

5. Cool the medium to 50–55 °C.

6. Under aseptic conditions, slowly add 10% sterile defibrinated sheep blood pre-warmed to 35 °C along the container wall.

7. Gently mix, pour into sterile Petri dishes, allow to solidify, and store appropriately.
