Formulation:​

  • Primary Stain: 0.3g basic fuchsin, 10ml 95% alcohol, 90ml 5% carbolic acid​

  • Decolorizing Solution: 3% hydrochloric acid in alcohol​

  • Counterstain: 0.3g methylene blue, 30ml 95% alcohol, 100ml 0.01% potassium hydroxide​

Intended Use:​

Specifically formulated for identifying acid-fast bacteria, enabling clear differentiation from other bacteria and cells under microscopy.​

How to Use:​

  1. Prepare a bacterial smear.​

  1. Fix the smear by passing through a flame.​

  1. Apply the primary stain, gently heat until steam appears, then remove from heat. Stain for 5 minutes (ensure the sample remains covered with stain by adding more if needed; avoid drying). After 5 minutes, rinse gently with distilled or deionized water and blot excess moisture with absorbent paper.​

  1. Apply the decolorizing solution repeatedly for 1-3 minutes until no visible red color remains. Rinse with water and blot dry.​

  1. Apply the counterstain and stain for 30-60 seconds. Rinse with water and blot excess moisture.​

  1. Microscopic examination: Acid-fast bacteria appear red against a light blue background; other bacteria and cells appear blue.
