Violet Red Bile Agar with Glucose and Lactose (VRB Agar) is a selective and differential solid culture medium designed for the isolation and enumeration of enteric bacteria, particularly coliform organisms, in food, water, and environmental samples.

The presence of bile salts and crystal violet inhibits Gram-positive bacteria, while lactose and glucose, in combination with neutral red, allow differentiation based on carbohydrate fermentation. Fermenting colonies typically appear red to purple with precipitation zones.

⚗️ Preparation Instructions

Pour Plate Method

  1. Suspend 50.1 g of dehydrated medium in 1,000 ml of distilled water
  2. Heat with continuous stirring and boil for no more than 1 minute
  3. Dispense into flasks and autoclave at 121 °C for 15 minutes
  4. Cool to 45–50 °C
  5. Add 1 ml of prepared sample to the center of a sterile 9 cm Petri dish
  6. Pour approximately 15 ml of medium into the plate and mix gently
  7. Allow to solidify
  8. Incubate inverted at 36 ± 1 °C for 18–24 hours

Spread Plate Method

  1. Pour medium into sterile Petri dishes at 45–50 °C
  2. Allow agar to solidify completely
  3. Dry plates inverted at 37 °C for 1–2 hours
  4. Spread 1 ml of appropriately diluted sample onto the agar surface
  5. Incubate inverted at 36 ± 1 °C for 18–24 hours

🧫 Storage & Stability

  • Prepared plates should be used immediately
  • Avoid direct light exposure
  • Dehydrated medium must be stored dry, protected from light, at 2–8 °C

⚠️ Limitations & Invalid Use

  • Do not use dehydrated medium after expiration date
  • Do not use if caking, discoloration, or moisture absorption is observed
