This base medium requires post-sterilization addition of urea to accurately identify urease-producing bacteria through distinctive phenol red color transition.

⚠️ Critical Preparation:
△ Must add 20% sterile urea solution (4g/100ml)
△ Add at 50-55°C to prevent urea degradation
△ For trained microbiologists only

Technical Specifications:
▶ Base Formula (per liter):
Peptone 1.0g | K₂HPO₄ 2.0g
NaCl 5.0g | Dextrose 1.0g | Phenol Red 0.012g
▶ pH: 6.9±0.1 (25°C)
▶ Preparation:

  1. 9g/L → sterilize at 121°C/15min

  2. Cool 50-55°C → add 40g/L urea

Diagnostic Color Transition:

  • Positive: Deep pink/red (alkaline)

  • Negative: Yellow/orange (acidic)

Applications:

  • Identification of Proteus and Helicobacter

  • Differentiation of Enterobacteriaceae

  • Urinary pathogen screening
