Trypticase Soy Sheep Blood Agar Base (TSSB) is a specialized microbiological medium used for the hemolysis test of Bacillus cereus. When supplemented with 5% sterile defibrinated sheep blood, this medium provides reliable and consistent results in bacterial hemolysis detection for laboratory and research applications.

Medium Composition (per liter):

  • Tryptone (Pancreatic Digest of Casein) 15.0g

  • Enzymatic Digest of Soybean Meal 5.0g

  • Sodium Chloride 5.0g

  • Agar 15.0g

Preparation Instructions:

  1. Dissolve 40.0g of the medium in 1L of distilled or deionized water.

  2. Heat to boiling until completely dissolved.

  3. Dispense into suitable containers.

  4. Sterilize by autoclaving at 121°C for 15 minutes.

  5. Mix well after sterilization to prevent agar sedimentation.

  6. Keep the medium in a water bath at 45–50°C.

  7. Add 5mL sterile defibrinated sheep blood per 100mL medium, mix gently, and pour into plates.

  8. Allow to solidify before use.

This TSSB medium ensures consistent performance in Bacillus cereus hemolysis tests, making it widely applicable in microbiological research, food safety testing, and laboratory quality control.
