Formula Composition (per liter):
– Tryptone: 17.0g
– Enzymatic Digest of Soybean Meal: 3.0g
– Dipotassium Phosphate: 2.5g
– Sodium Chloride: 5.0g
– Dextrose: 2.5g
– Final pH: 7.3 ± 0.2 (25°C)
Preparation Protocol:
Standard TSB:
1. Suspend 30.0g in 1L distilled water
2. Warm to 45°C with stirring until dissolved
3. Autoclave at 121°C for 15 minutes
Double-Strength TSB:
– Use 60.0g per liter
Modified TSB (mTSB) Preparation:
1. Prepare and sterilize base broth
2. Cool to ≤50°C
3. Aseptically add per 225ml:
– Polymyxin B (2.25mg)
– Nalidixic Acid (2.25mg)
