Triple Sugar Iron Agar Medium (TSI Agar) is a dehydrated microbiological culture medium designed for biochemical differentiation of Enterobacteriaceae. The medium is widely used in microbiology laboratories for carbohydrate utilization and hydrogen sulfide production testing.

Medium Composition (per liter)

  • Pancreatic Digest of Casein: 10.0 g

  • Peptic Digest of Animal Tissue: 10.0 g

  • Sodium Chloride: 5.0 g

  • Lactose: 10.0 g

  • Sucrose: 10.0 g

  • Dextrose: 1.0 g

  • Ferric Ammonium Citrate: 0.2 g

  • Sodium Thiosulfate: 0.2 g

  • Phenol Red: 0.025 g

  • Agar: 13.0 g

Final pH: 7.3 ± 0.2 (at 25 °C)


Preparation Instructions

Suspend 59.4 g of the medium in 1 liter of distilled or deionized water. Heat with agitation and boil until completely dissolved. Dispense into test tubes and sterilize by autoclaving at 121 °C for 15 minutes. After sterilization, allow the medium to solidify in a deep slant position before use.
