Medium Composition (per liter):

• Casamino Acids (Acid Hydrolyzed Casein) 10.0 g

• Enzymatic Digest of Soybean Meal (Soy Peptone) 5.0 g

• Yeast Extract 2.0 g

• Glucose 5.0 g

• L-Cysteine Hydrochloride 0.5 g

• K₂HPO₄·7H₂O 2.0 g

• MgCl₂·6H₂O 0.5 g

• ZnSO₄·7H₂O 0.25 g

• CaCl₂ 0.15 g

• FeCl₃ 0.1 mg

• Tween 80 1.0 ml

• Agar 20.0 g

Preparation Instructions:

1. Weigh 46.4 g of the medium and suspend in 1L distilled or deionized water (adjust proportionally if preparing different volumes).

2. Heat to boiling until completely dissolved.

3. Dispense into appropriate containers.

4. Sterilize by autoclaving at 121°C for 15 minutes.

5. Shake gently after sterilization to prevent agar settling at the bottom before solidification.

⚠️ Note: A small amount of insoluble material may be present, which does not affect the medium’s performance.
