
Toluidine Blue DNase Agar Powder for Thermostable Nuclease Test
Toluidine Blue DNase Agar is a dehydrated culture medium used for thermostable nuclease testing.
It is supplied as a dry powder, packaged in 100g bottles, with a shelf life of 3 years at room temperature.
Composition per liter:
Deoxyribonucleic Acid (DNA): 0.3 g
Calcium Chloride (anhydrous): 1.1 mg
Sodium Chloride: 10.0 g
Toluidine Blue O: 0.083 g
Tris (Tris(hydroxymethyl)aminomethane): 6.1 g
Agar: 10.0 g
Final pH: 9.0 ± 0.2 (at 25°C)
Preparation:
Suspend 26.5 g of the powder in 1 L of distilled water. Heat to boiling for 1 minute until completely dissolved. Cool to about 50°C, pour into plates, and allow to solidify. Use on the same day.
If not used immediately, sterilize by autoclaving at 116°C for 20 minutes.
