Todd-Hewitt Broth is widely used in clinical and research microbiology for cultivating streptococci and preparing beta-hemolysin. Its balanced nutrient composition promotes rapid growth while maintaining the desired physiological properties of the bacteria.

Composition (per liter):

  • Peptone – 20.0 g

  • Heart Extract Powder – 3.1 g

  • Dextrose – 2.0 g

  • Sodium Chloride – 2.0 g

  • Disodium Hydrogen Phosphate – 0.4 g

  • Sodium Carbonate – 2.5 g

  • Final pH: 7.8 ± 0.1 (at 25℃)

Preparation Instructions:

  1. Dissolve 30.0 g of dehydrated medium in 1 liter of distilled or deionized water (adjust proportionally for other volumes).

  2. Heat to boiling until completely dissolved.

  3. Dispense into suitable containers.

  4. Sterilize by autoclaving at 121℃ for 15 minutes.

  5. Cool and store under aseptic conditions until use.

Storage:
Store in a tightly sealed container at room temperature in a dry place. Shelf life: 3 years.
