Terrific Broth (TB Medium) is a specialized microbial growth medium tailored for E. coli cultivation. Its formulation ensures prolonged bacterial growth, making it a preferred choice for molecular biology experiments and industrial-scale protein production.
Formula (Per Liter):
Pancreatic Digest of Casein (Trypticase Peptone): 12.0g
Yeast Extract: 24.0g
Dipotassium Phosphate (K₂HPO₄): 9.4g
Monopotassium Phosphate (KH₂PO₄): 2.2g
pH: 7.2 ± 0.2 (at 25°C)
Usage Instructions
Weigh 47.6g of the powder into 1L of distilled or deionized water.
Add 4ml glycerol and mix until fully dissolved.
Sterilize by autoclaving at 121°C for 15 minutes.
Store in a dark, cool place after preparation.
Storage & Shelf Life
Storage: Store unopened bottles at room temperature (15–25°C).
Shelf Life: 3 years from the date of manufacture.
