Preparation Instructions

  1. Dissolve 34.6 g of the agar base in 1 L of distilled or deionized water.

  2. Boil for more than 1 minute to ensure complete dissolution.

  3. Sterilize by autoclaving at 121℃ for 15 minutes.

  4. Shake well after sterilization to prevent agar sedimentation.

  5. Cool to 50–55℃.

  6. Aseptically add per 100 ml medium: 5 ml of 20% sterilized sugar solution and 2 vials of 50% egg-yolk saline suspension (10 ml).

  7. Mix gently and pour into plates or tubes. Allow to solidify before use.
