Staphylococcus Enrichment Broth Base – Selective Medium for Coagulase-Positive Staphylococci (250g)

Staphylococcus Enrichment Broth Base is a selective liquid medium designed for the enrichment of coagulase-positive Staphylococcus species, including Staphylococcus aureus.

Its formulation supports optimal growth of target organisms while inhibiting competing flora, making it ideal for use in food microbiology, clinical diagnostics, and research laboratories.

The combination of peptones, mannitol, glycine, lithium chloride, and sodium pyruvate provides a nutrient-rich and selective environment that enhances recovery of pathogenic staphylococci before confirmatory testing.

Composition (per liter):

• Soya Peptone – 3.5 g

• Yeast Extract – 6.5 g

• Trypticase Peptone – 10.0 g

• Mannitol – 5.0 g

• Glycine – 10.0 g

• Dipotassium Phosphate – 5.0 g

• Lithium Chloride – 5.0 g

• Sodium Pyruvate – 10.0 g

• Final pH: 7.3 ± 0.1 (at 25°C)

Preparation & Instructions:

1. Suspend 55.0 g of the medium in 1 liter of distilled or deionized water.

2. Heat with stirring until completely dissolved.

3. Dispense into appropriate containers.

4. Sterilize by autoclaving at 121°C for 15 minutes.

5. Allow to cool to room temperature.

6. Under aseptic conditions, add 1 vial of supplement S3404 per 100 ml medium.

7. Mix well and use immediately.
