Intended Use:
Designed for staining bacterial spores, facilitating the visualization and identification of spores in laboratory settings.
How to Use:
Smear Preparation: Prepare a bacterial smear (avoid excessive density), air-dry, and fix.
Staining: Apply 3-5 drops of malachite green stain to cover the smear. Use tweezers or a test tube holder to hold the slide, and heat over a flame for approximately 5 minutes (start timing when the stain begins to steam). Add more malachite green stain as needed during heating; do not allow boiling or drying of the specimen.
Rinsing: Once the slide has cooled, rinse with distilled water.
Counterstaining: Apply safranin stain to cover the smear, counterstain for 1 minute, then rinse with water and blot dry.
Microscopy: Observe under an oil immersion lens.
Result Interpretation:
Spores appear green.
Bacterial vegetative cells appear red.
