Sauton Broth Medium Base is a liquid culture medium designed for the cultivation of Bacillus Calmette–Guérin (BCG) and the preparation of Purified Protein Derivative (PPD). It can also be used to prepare Sauton Potato Medium and diluted Sauton Medium. The medium requires supplementation with glycerol to support optimal mycobacterial growth.

Composition (per liter)

  • Asparagine – 4.0g

  • Citric Acid – 2.0g

  • Dipotassium Phosphate – 0.5g

  • Magnesium Sulfate Heptahydrate – 0.5g

  • Ferric Ammonium Citrate – 0.05g

  • pH 7.0 ± 0.2

Preparation Instructions

  1. Dissolve 7.0g of Sauton Broth Medium Base in 940ml distilled or deionized water.

  2. Add 60ml glycerol and adjust pH to 7.0 ± 0.2 using NaOH.

  3. Heat to boiling to completely dissolve the powder.

  4. Dispense and sterilize at 121℃ for 15 minutes.

  5. Cool and store until use.

Notes

  • The medium is hygroscopic and may form clumps; this does not affect performance.

  • Ensure proper aseptic technique during preparation and supplementation.
