S Medium – Agar Medium for Actinomycete Cultivation, 250g

S Medium is a specialized culture medium designed for the isolation and cultivation of actinomycetes. Its balanced formulation of carbon, nitrogen, minerals, and trace elements supports consistent growth and differentiation of actinomycete species, making it suitable for microbiology research and industrial applications.

Complete Formula (per liter):

• Glucose: 10.0 g

• Potassium Nitrate (KNO₃): 1.0 g

• Magnesium Sulfate·7H₂O (MgSO₄·7H₂O): 0.1 g

• Calcium Chloride·2H₂O (CaCl₂·2H₂O): 0.1 g

• Dipotassium Phosphate (K₂HPO₄): 0.5 g

• Trace Element Solution: 1.0 ml

• Ferric Citrate Stock Solution: 1.0 ml

• Agar: 20.0 g

• Final pH: 7.2 ± 0.2 at 25°C

Instructions for Use:

1. Weigh 33.7 g of the medium and suspend in 1 liter of distilled or deionized water.

2. Heat to boiling for at least 1 minute to ensure complete dissolution.

3. Dispense into containers.

4. Sterilize at 116°C for 30 minutes.

5. Mix gently after sterilization, pour into plates, allow to solidify, and store for later use.
