Medium Composition (per liter)

  • Tryptone – 10.0 g

  • Yeast Extract – 5.0 g

  • Dextrose – 10.0 g

  • Arabinose – 5.0 g

  • Sucrose – 5.0 g

  • Sodium Acetate – 15.0 g

  • Monopotassium Phosphate – 6.0 g

  • Ammonium Citrate – 2.0 g

  • Magnesium Sulfate – 0.57 g

  • Manganese Sulfate – 0.12 g

  • Ferrous Sulfate – 0.03 g

  • Agar – 15.0 g

  • pH: 5.4 ± 0.2 (25℃)

Preparation Instructions

  1. Dissolve 73.7 g of the medium in 1 liter of distilled or deionized water.

  2. Boil for 1 minute until fully dissolved.

  3. Add 1 ml Tween 80 and 1.32 ml glacial acetic acid, mix thoroughly.

  4. Adjust pH to 5.4 ± 0.2 (25℃).

  5. Continue boiling for 2–3 minutes. Do not autoclave.

  6. Cool and pour plates for use.
