Medium Composition
Composition (per liter)
Histidine — 0.5 g
Raffinose — 2.5 g
Dipotassium Phosphate Trihydrate (K2HPO4·3H2O) — 1.0 g
Magnesium Sulfate Heptahydrate (MgSO4·7H2O) — 0.5 g
Ferrous Sulfate Heptahydrate (FeSO4·7H2O) — 0.01 g
Calcium Chloride — 0.02 g
Agar — 20.0 g
Final pH: 7.2 ± 0.2 (25°C)
Directions for Use
- Suspend 24.5 g of the medium in 1 liter of distilled or deionized water.
- Heat to boiling for approximately 1 minute until completely dissolved.
- Dispense the medium into appropriate containers.
- Sterilize by autoclaving at 121°C for 15 minutes.
- Allow the medium to cool and pour into sterile petri dishes if agar plates are required.
