Medium Composition (per liter):

• Peptone 20.0 g

• Dextrose 5.0 g

• Yeast Extract 10.0 g

• L-Cysteine Hydrochloride 0.5 g

• Calcium Chloride 0.008 g

• Magnesium Sulfate 0.008 g

• Dipotassium Phosphate 0.04 g

• Monopotassium Phosphate 0.04 g

• Sodium Bicarbonate 0.4 g

• Sodium Chloride 0.08 g

Preparation Instructions:

1. Dissolve 36 g of the medium in 1L of distilled or deionized water.

2. Heat until completely dissolved.

3. Dispense into containers.

4. Sterilize by autoclaving at 121°C for 15 minutes.

5. Cool to below 50°C, then add supplements:

   • 1 vial Hematin Chloride Solution (0.5 mg) per 200 ml medium

   • 2 vials Vitamin K1 Solution (0.2 mg) per 200 ml medium

6. Mix thoroughly before use.
