Medium Composition (per liter)
Beef heart infusion solids (from 500 g beef heart): 13.0 g
Tryptose: 10.0 g
Sodium Chloride: 5.0 g
Ethylenediaminetetraacetic acid (EDTA): 0.3 g
Agar: 15.0 g
Final pH: 7.3 ± 0.2 (25℃)
Preparation Instructions
Suspend 43.3 g of the medium in 1 L distilled or deionized water.
Heat to boiling until completely dissolved.
Dispense into suitable containers and autoclave at 121℃ for 15 minutes.
Shake well after sterilization to avoid agar settling at the bottom.
Cool to 55℃, then aseptically add (per 100 ml medium):
4 mg thallium acetate
30,000 U lysozyme
3,000 U polymyxin B
Mix thoroughly and prepare plates for use.
