PLET Agar Base is a selective culture medium designed for the isolation of Bacillus anthracis. When supplemented with thallium acetate, lysozyme, and polymyxin B, this medium inhibits the growth of most contaminating microorganisms, providing a reliable platform for the selective recovery and differentiation of anthrax bacilli.

Medium Composition (per liter)

  • Beef heart infusion solids (from 500 g beef heart): 13.0 g

  • Tryptose: 10.0 g

  • Sodium Chloride: 5.0 g

  • Ethylenediaminetetraacetic acid (EDTA): 0.3 g

  • Agar: 15.0 g

  • Final pH: 7.3 ± 0.2 (25℃)

Preparation Instructions

  1. Suspend 43.3 g of the medium in 1 L distilled or deionized water.

  2. Heat to boiling until completely dissolved.

  3. Dispense into suitable containers and autoclave at 121℃ for 15 minutes.

  4. Shake well after sterilization to avoid agar settling at the bottom.

  5. Cool to 55℃, then aseptically add (per 100 ml medium):

    • 4 mg thallium acetate

    • 30,000 U lysozyme

    • 3,000 U polymyxin B

  6. Mix thoroughly and prepare plates for use.
