Pick’s Broth Base – Enrichment Medium for Hemolytic Streptococci, 100g

Pick’s Broth Base is a specialized microbiological medium used for the enrichment and cultivation of hemolytic streptococci. Its composition supports selective growth while inhibiting unwanted flora, making it suitable for laboratory diagnostics and research.

Composition (per liter):

• Tryptone  – 10.0 g

• Heart Extract Powder  – 20.0 g

• Crystal Violet  – 0.002 g

• Final pH: 7.4 ± 0.1 (at 25℃)

Preparation Instructions:

1. Suspend 30.0 g of Pick’s Broth Base in 1 liter of distilled or deionized water.

2. Warm gently to dissolve.

3. Dispense into suitable containers.

4. Sterilize by autoclaving at 121℃ for 15 minutes.

5. Cool to 50–55℃.

6. Under aseptic conditions, supplement each 50 ml of basal medium with:

   • 1 ampoule of 1:800 sodium azide solution

   • 2.5 ml of sterile defibrinated sheep blood

7. Mix gently and store for use.

Storage: Store tightly sealed in a cool, dry place at room temperature. Shelf life is 3 years.
