O/F Medium (HLGB) – 250g | Glucose Utilization Test for Vibrio Identification

O/F Medium (HLGB) is designed for testing glucose metabolism in Vibrio species through Oxidation/Fermentation (O/F) differentiation.

Formula (Per Liter):

– Peptone: 2.0g

– Yeast Extract: 0.5g

– Sodium Chloride: 30.0g

– Dextrose: 10.0g

– Bromcresol Purple: 0.015g

– Agar: 3.0g

pH 7.4 ± 0.2 (25°C)

Protocol:

1️⃣ Suspend 45.6g in 1L water

2️⃣ Boil 1 minute

3️⃣ Dispense into tubes

4️⃣ Autoclave at 121°C for 15 minutes

5️⃣ Solidify upright

Test Procedure:

– Inoculate two tubes

– Add sterile mineral oil to one tube

– Observe color changes
