Nodule Bacteria Medium (YM) is a specialized microbiological medium designed for the cultivation of Rhizobium and other nitrogen-fixing bacteria involved in legume root nodulation. This medium contains Congo Red to aid in the visualization and differentiation of bacterial colonies, making it an essential tool for studying symbiotic relationships between plants and bacteria. It provides all the necessary nutrients to support the growth and development of Rhizobium species.

Instructions for Use:
To prepare, dissolve 29.83g of Nodule Bacteria Medium in 1L of distilled or deionized water. Boil until fully dissolved. Sterilize by autoclaving at 121°C for 30 minutes. After sterilization, gently shake the medium to mix and prevent agar from settling at the bottom. Cool and store until use.

Storage: Store at room temperature, away from direct sunlight. Shelf life: 3 years.

  • Dipotassium Phosphate (K2HPO4): 0.5g

  • Magnesium Sulfate (MgSO4): 0.2g

  • Sodium Chloride (NaCl): 0.1g

  • Mannitol: 10.0g

  • Yeast Extract: 1.0g

  • Congo Red: 0.025g

  • Agar: 18.0g

  • pH: 7.0 ± 0.2 at 25°C
