Peptone — 20.0 g
Nalidixic Acid — 0.015 g
Magnesium Sulfate — 0.3 g

Principle
Peptone provides nutrients required for microbial growth. Magnesium sulfate and dipotassium phosphate supply essential ions in the culture medium. Cetrimide acts as a selective agent that suppresses competing microorganisms. Nalidixic acid functions as a selective antimicrobial component.
Preparation Instructions
- Suspend 35 g of the medium in 1000 ml purified water.
- Heat the mixture until the medium is completely dissolved.
- Dispense the medium into appropriate containers.
- Sterilize by autoclaving at 121°C for 15 minutes.
- Allow the medium to cool before use.
Note:
Because the medium contains magnesium sulfate, the sterilized medium may appear slightly turbid and small amounts of sediment may be present.
