🧾 Medium Composition (per liter)

Letheen Agar Base Components

• Beef Extract – 3.0 g

• Pancreatic Digest of Casein – 5.0 g

• Dextrose – 1.0 g

• Lecithin – 1.0 g

• Agar – 15.0 g

Additional Modified Components

• Tryptone – 5.0 g

• Proteose Peptone – 10.0 g

• Yeast Extract – 2.0 g

• Sodium Chloride – 5.0 g

• Sodium Bisulfite – 0.1 g

• Agar – 5.0 g

Final pH: 7.2 ± 0.2 (25 °C)

⚙️ Directions for Use

1. Suspend 52.1 g of the medium in 1 liter of distilled or deionized water.

2. Heat and boil for at least 1 minute until agar is completely dissolved.

3. Add 1 unit of Polysorbate 80 (Tween 80) per liter, or 1 unit per 100 mL as required.

4. Mix thoroughly and dispense into suitable containers.

5. Sterilize by autoclaving at 121 °C for 15 minutes.

6. Shake well immediately after sterilization.

7. Pour into sterile Petri dishes, allow to solidify, and prepare for use.
