Modified Giolitti-Cantoni Broth Base Powder for Staphylococcus MPN

Modified Giolitti-Cantoni Broth Base is a dehydrated culture medium designed for the Most Probable Number (MPN) method of staphylococcus enumeration.
It is provided in dry powder form, packed in 250g bottles, with a shelf life of 3 years when stored at room temperature.

Composition per liter:

• Tryptone (10.0 g)

• Beef Extract (5.0 g)

• Yeast Extract (5.0 g)

• D-Mannitol (20.0 g)

• Sodium Chloride (5.0 g)

• Lithium Chloride (5.0 g)

• Glycine (1.2 g)

• Sodium Pyruvate (3.0 g)

• Tween 80 (1.0 g)

• Final pH: 6.9 ± 0.2 (at 25°C)

Preparation:
Dissolve 55.2 g of powder in 1 L distilled or deionized water, sterilize at 121°C for 15 minutes. Cool to about 50°C, then aseptically add 1.05 ml of 1% potassium tellurite solution and mix well. Dispense 19 ml per sterile test tube (20 × 200 mm).
For double-strength medium, use 110.4 g per liter and adjust pH before sterilization.
