Modified Giolitti-Cantoni Broth Base
Modified Giolitti-Cantoni Broth Base is a microbiological culture medium formulated for the enumeration of Staphylococcus species using the Most Probable Number (MPN) method.
The modified formulation includes Tween 80, which enhances microbial recovery and improves performance in selective enrichment procedures. When supplemented with potassium tellurite solution, the medium becomes suitable for selective growth of Staphylococcus species.

Preparation Instructions
- Suspend 55.2 g of the medium in 1 liter of distilled or deionized water.
- Sterilize by autoclaving at 121°C for 15 minutes.
- Cool the medium to approximately 50°C.
- Under aseptic conditions, add 1.05 ml of 1% potassium tellurite solution and mix well.
- Dispense 19 ml per sterile test tube (20 × 200 mm) for use.
Preparation of Double-Strength Medium
For double-strength formulation, dissolve 110.4 g of the medium in 1 liter of water, adjust pH if necessary, and sterilize under the same conditions.
Storage
Store tightly sealed in a cool and dry place at room temperature.
