Mitis Salivarius Broth Base is specially formulated for the selective cultivation and enrichment of Streptococcus mitis and related species from clinical or oral samples.
The medium contains pancreatic digest of casein and proteose peptone as nitrogen sources, phosphate buffer to maintain pH stability, and high sucrose concentration for enhanced differentiation of streptococci. The addition of potassium tellurite solution provides selectivity by suppressing non-target organisms.

  • Pancreatic digest of casein ……………… 15.0 g

  • Proteose peptone ……………………….. 5.0 g

  • Dipotassium phosphate ……………………. 4.0 g

  • Dextrose (Glucose) ………………………. 1.0 g

  • Sucrose ………………………………… 50.0 g

  • Trypan Blue ……………………………. 75 mg

  • Crystal Violet ………………………….. 0.8 mg

  • Final pH: 7.0 ± 0.2 (at 25℃)


📌 Instructions for Use

  1. Suspend 75.0 g of the medium in 1 liter of distilled or deionized water.

  2. Heat gently to dissolve completely.

  3. Dispense into appropriate containers.

  4. Sterilize by autoclaving at 121℃ for 15 minutes.

  5. Cool to 50–55℃.

  6. Under aseptic conditions, add 1 ampoule of potassium tellurite solution (2 mg) per 200 ml of medium.

  7. Mix thoroughly, dispense, and use as required.
