Composition (per liter)

• Ammonium Sulfate – 0.5g

• L-Glutamic Acid – 0.5g

• Sodium Citrate – 0.1g

• Pyridoxine – 1.0mg

• Biotin – 0.5mg

• Disodium Phosphate – 2.5g

• Monopotassium Phosphate – 1.0g

• Ferric Ammonium Citrate – 0.04g

• Magnesium Sulfate – 0.05g

• Calcium Chloride – 0.5mg

• Zinc Sulfate – 1.0mg

• Copper Sulfate – 1.0mg

• pH: 6.6 ± 0.2 (25℃)

Preparation Instructions

1. Dissolve 4.7g of the medium in 900ml distilled or deionized water.

2. Optional: add 2ml glycerol and 0.5g Tween-80 for enhanced growth.

3. Heat and boil to completely dissolve.

4. Dispense and sterilize at 121℃ for 10 minutes.

5. Cool to approximately 50℃, then aseptically add Middlebrook ADC enrichment (2 vials per 90ml medium) and mix gently.

6. Medium is ready for Mycobacterium cultivation.
