Application

Medium S1 is designed for the determination of nisin potency in microbiological testing. It provides a balanced nutrient composition to support reliable assay performance.

Composition (per liter)

• Tryptone – 8.0 g

• Yeast Extract – 5.0 g

• Dextrose – 5.0 g

• Sodium Chloride – 5.0 g

• Disodium Hydrogen Phosphate – 2.0 g

• Agar – 15.0 g

• pH 6.9 ± 0.1 (at 25℃)

Instructions for Use

1. Dissolve 40.0 g of the medium in 1 L of distilled or deionized water.

2. Heat to boiling for more than 1 minute until completely dissolved.

3. Dispense 200 ml per container and sterilize at 121℃ for 20 minutes.

4. Shake after sterilization to avoid agar sedimentation.

5. Cool to about 70℃, then aseptically add one ampoule of 50% Tween 20 solution (Tween 20: water = 1:1) per 200 ml of medium.

6. Mix gently and prepare for use.
