Preparation Instructions

1. Weigh 54.0 g of the medium powder.
2. Dissolve in 1000 mL distilled or deionized water.
3. Heat and boil for at least 1 minute to completely dissolve the medium.
4. Dispense into suitable containers.
5. Sterilize at 115°C for 20 minutes.
6. After sterilization, mix well to prevent agar sedimentation and cool to 50–55°C.
7. Under aseptic conditions, add the following to each 200 mL of medium:

• Two vials of Polymyxin B solution (25,000 units each)
• One vial of Gentamicin solution

8. Mix gently and pour into sterile Petri dishes.
