M17 Agar is a high-quality medium designed for the detection and enumeration of lactic acid bacteria in milk and dairy products. This medium requires supplementation with 10% lactose solution before use and is widely used in dairy microbiology laboratories.

Composition (per liter):

  • Tryptic Digest of Casein – 2.5 g

  • Peptic Digest of Meat – 2.5 g

  • Papain Digest of Soya – 5.0 g

  • Yeast Extract – 2.5 g

  • Meat Extract – 5.0 g

  • β-Glycerophosphate (disodium salt) – 19.0 g

  • Ascorbic Acid – 0.5 g

  • Magnesium Sulfate Heptahydrate – 0.25 g

  • Agar – 11.0 g

  • pH: 6.9 ± 0.2 (25℃)

Preparation Instructions:

  1. Dissolve 4.83 g of the powder in 95 mL distilled or deionized water, heat and boil until fully dissolved.

  2. Dispense and sterilize by autoclaving at 121℃ for 15 minutes.

  3. Cool to 55℃, then aseptically add 5 mL sterile 10% lactose solution per 100 mL medium and mix gently.

  4. Pour plates and allow to solidify; ready for detection of lactic acid bacteria in milk and dairy products.
