M1 Medium – Agar-Based Medium for Actinomycete Cultivation, 250g

M1 Medium is a selective culture medium formulated for the isolation and cultivation of Actinomycetes. Enriched with L-Asparagine, yeast extract, nitrate, betaine, and agar, it provides essential nutrients and osmoprotectants to support the stable growth and differentiation of actinomycete strains.

Complete Formula (per liter):

• L-Asparagine: 1.0 g

• Yeast Extract: 5.0 g

• Potassium Nitrate (KNO₃): 5.0 g

• Betaine: 1.25 g

• Agar: 12.0 g

• Final pH: 7.2 ± 0.2 at 25°C

Instructions for Use:

1. Dissolve 24.3 g of the medium in 1 liter of distilled or deionized water.

2. Add 10 vials of glycerol to enrich the medium.

3. Heat to boiling for at least 1 minute until completely dissolved.

4. Dispense into containers and sterilize at 121°C for 15 minutes.

5. Shake thoroughly after sterilization.

6. Cool to 50–55°C, pour into plates, allow to solidify, and use as required.
