Selective Culture Medium for Detection and Enumeration of Pseudomonas aeruginosa in Water

m-Pseudomonas aeruginosa Agar (m-PA Agar) is a microbiological culture medium designed for the detection and enumeration of Pseudomonas aeruginosa in water samples using the membrane filtration method.

The medium supports bacterial growth and enables the identification of characteristic colonies when used with selective supplements. It is commonly applied in drinking water testing, environmental water monitoring, swimming pool water analysis, and microbiological quality control laboratories.

Preparation Instructions

  1. Weigh 39.7 g of the medium powder.
  2. Dissolve in 1000 mL distilled or deionized water.
  3. Heat and boil for at least 1 minute to completely dissolve the medium.
  4. Dispense into suitable containers.
  5. Sterilize at 121°C for 15 minutes.
  6. After sterilization, mix well to prevent agar sedimentation and cool to 50–55°C.
  7. Under aseptic conditions, add the following supplements to each 100 mL of medium:
  • Sodium sulfapyridine solution
  • m-PA agar supplement
  • Nalidixic acid solution
  • Kanamycin solution
  1. Mix gently and pour into sterile Petri dishes.
