⚗️ Medium Preparation (General Guidelines)
1️⃣ Dissolution
Add the required amount of dehydrated medium to distilled or deionized water
Allow to stand for 10–15 minutes to facilitate hydration
Stir, shake, or swirl gently to promote dissolution
If necessary, heat gently with constant stirring until fully dissolved
Avoid excessive heating or prolonged high temperatures to prevent nutrient degradation
2️⃣ pH Adjustment
The pH of the medium should be measured at 25 ± 2 °C
Adjust pH using sterile solutions if required:
1 mol/L sodium hydroxide
10% sodium carbonate
1 mol/L hydrochloric acid
Note: The pH after sterilization may differ slightly from pre-sterilization values.
It is common practice to adjust the pH approximately 0.2 units higher before autoclaving
Although dehydrated media are pre-adjusted, pH verification before use is recommended
3️⃣ Clarification
After pH adjustment, heat and filter if necessary to obtain a clear medium
