Composition (per liter)

  • Asparagine – 3.6g

  • Monopotassium Phosphate – 2.4g

  • Magnesium Sulfate – 0.24g

  • Magnesium Citrate – 0.6g

  • Potato Flour – 30.0g

  • Malachite Green – 0.4g

Preparation Instructions

  1. Dissolve 37.4g of the medium in 600ml distilled or deionized water containing 12ml glycerol.

  2. Sterilize at 121℃ for 15 minutes.

  3. Cool to approximately 50℃.

  4. Aseptically add 1000ml whole egg and mix gently, avoiding foam formation.

  5. Let stand for 1 hour, then dispense into tubes.

  6. Place tubes at an angle in a serum coagulator and gently heat at 80–85℃ for 1–1.5 hours to solidify the slant.

  7. Medium is ready for Mycobacterium isolation and cultivation.
