Lowenstein-Jensen Medium Base is specifically formulated for the selective isolation and identification of Mycobacterium species, including Mycobacterium tuberculosis. The medium contains asparagine, potato flour, and malachite green to support optimal growth while suppressing contaminants.

Asparagine – 3.6g
Monopotassium Phosphate – 2.4g
Magnesium Sulfate – 0.24g
Magnesium Citrate – 0.6g
Potato Flour – 30.0g
Malachite Green – 0.4g
Preparation Instructions
Dissolve 37.4g of the medium in 600ml distilled or deionized water containing 12ml glycerol.
Sterilize at 121℃ for 15 minutes.
Cool to approximately 50℃.
Aseptically add 1000ml whole egg and mix gently, avoiding foam formation.
Let stand for 1 hour, then dispense into tubes.
Place tubes at an angle in a serum coagulator and gently heat at 80–85℃ for 1–1.5 hours to solidify the slant.
Medium is ready for Mycobacterium isolation and cultivation.
