Formula Composition (per liter):

– Beef Extract: 5.0g

– Proteose Peptone No.3: 10.0g

– Sodium Chloride: 5.0g

– Lecithin: 0.7g

– Final pH: 7.0 ± 0.2 (25°C)

Preparation Protocol:

1. Suspend 20.7g in 1L distilled water

2. Heat to 80°C with stirring until dissolved

3. Add 1g polysorbate 80 per 200ml broth

4. Autoclave at 121°C for 15 minutes

For viscous products: Pre-dilute 1:10 in sterile saline

Technical Note:

– Lecithin may cause initial cloudiness – clears upon sterilization

– Optimal incubation: 30-35°C for 48-72 hours
