Composition (per liter)

  • Casein enzymic hydrolysate – 10.0g

  • Yeast extract – 10.0g

  • Lactose – 5.0g

  • Sucrose – 5.0g

  • Calcium carbonate – 3.0g

  • Dipotassium phosphate – 0.5g

  • Bromocresol purple – 0.02g

  • Agar – 18.0g

  • pH: 7.0 ± 0.2 (25℃)

Preparation Instructions

  1. Dissolve 51.52g of the medium in 1L of distilled or deionized water.

  2. Heat and boil for at least 1 minute to fully dissolve.

  3. Dispense into containers and sterilize at 121℃ for 15 minutes.

  4. Cool the medium to approximately 50℃ before preparing plates.

  5. Gently shake to evenly distribute calcium carbonate insoluble particles throughout the plates.

  6. Plates are ready for use in bacterial enumeration.
