Korthof Medium Base for Leptospira Enrichment Culture – 250g

Korthof Medium Base is a specialized microbiological medium designed for the enrichment and cultivation of Leptospira species. This medium requires supplementation with sterile rabbit serum to support the growth of fastidious leptospires. It is ideal for clinical microbiology, research laboratories, and studies on pathogenic spirochetes.

Instructions for Use:

1. Dissolve 3.42g of Korthof Medium Base in 100mL of distilled or deionized water. Boil until completely dissolved.

2. Sterilize by autoclaving at 121°C for 20 minutes.

3. Cool to 50–55°C. Under sterile conditions, add 8–10mL sterile rabbit serum per 100mL medium. Mix gently.

4. Dispense 5mL per sterile tube and incubate in a 56°C water bath for 30 minutes.

5. Optionally, to prevent contamination, sodium sulfadiazine can be added at 0.5mg/mL. Medium is now ready for enrichment culture.

Storage: Store at room temperature, away from direct sunlight. Shelf life: 3 years.

• Sodium Chloride (NaCl): 14.0g/L

• Sodium Bicarbonate (NaHCO3): 0.2g/L

• Potassium Chloride (KCl): 0.4g/L

• Calcium Chloride (CaCl2): 0.4g/L

• Potassium Dihydrogen Phosphate (KH2PO4): 2.4g/L

• Disodium Phosphate (Na2HPO4): 8.8g/L

• pH: 7.2 ± 0.2 at 25°C

• Supplement (not included): Sterile rabbit serum, optional sodium sulfadiazine
