Hoyle Medium Base

Hoyle Medium Base is formulated for the selective isolation and cultivation of Corynebacterium diphtheriae (diphtheria bacillus). This medium base must be supplemented with sterile defibrinated sheep blood and potassium tellurite solution for optimal bacterial growth and selectivity.

Specifications:

• Form: Powder, 250 g/bottle

• Storage: Room temperature

• Shelf Life: 3 years

Preparation Instructions:

1. Dissolve the powder in distilled or deionized water as required.

2. Dispense into containers or plates.

3. Sterilize by autoclaving at 121℃ for 15 minutes.

4. Cool to 50–55℃.

5. For use, supplement each 100 ml of sterilized medium with:

   • 5 ml Sterile Defibrinated Sheep Blood

   • 1 unit (35 mg) of 3.5% Potassium Tellurite solution

6. Mix gently under aseptic conditions and proceed with inoculation.
