GYM Agar (Glucose Yeast Malt Agar) is a nutrient-rich culture medium widely used for the cultivation of actinomycetes, Streptomyces, and related microorganisms. The balanced composition of glucose, yeast extract, and malt extract provides essential carbon, nitrogen, and growth factors. The addition of calcium carbonate helps buffer the medium and supports microbial development.

Complete Formula (per liter):

  • Glucose: 4.0 g

  • Yeast Extract: 4.0 g

  • Malt Extract: 10.0 g

  • Calcium Carbonate: 2.0 g

  • Agar: 12.0 g

  • Final pH: 7.2 ± 0.2 at 25°C

Instructions for Use:

  1. Suspend 32.0 g of the medium in 1 liter of distilled or deionized water.

  2. Heat to boiling and maintain for at least 1 minute until agar is completely dissolved.

  3. Stir and dispense into containers.

  4. Sterilize at 121°C for 15 minutes.

  5. Cool to 55–60°C, mix gently, pour into plates, allow to solidify, and store for use.
